Agrobacteium - mediated Transformation of Korean Lawngrass (Zoysia japonica)

Abstract
Embryogenic calli were induced from hulled seeds of zoysiagrass cv. Zenith. One callus line Z-5 derived from single seed with fast growing character and high regeneration rate was selected for transformation experiment. A cointegrate vector pTOK 233, harboring HPT (hygromycin phosphotransferase) and GUS (¥â-glucuronidase) genes in Agrobacterium LBA 4404 was used. Transient GUS reaction in callus was detected and its expression was promoted with sonication and vacuum treatments. Callus selection was done on MS medium in the presence of 40¡­50 §·¡¤L^(-1) hygromycin for 6 months. Newly-formed callus from this selection regenerated into green shoot clusters and plantlets, and also GUS expression was detected in the green shoot clusters. In this paper we report the establishment of an efficient Agrobacterium-mediated transformation system for zoysiagrass for the first time.
